This may be a dumb question, but is it any more difficult to build a left-hander?

No - building a leftie isn't really any harder for the most bit
It is a little different  when it comes to setting the guitar up as you have to rely upon careful measurements alone (we carefully measure on right handers too), but as I'm not a natural leftie I cant "road test" the guitar in the same way that I might a right handed guitar.
