If you don't split it does not matter. However, if you later decide you want to split, it could.

Keep in mind you could get interresting splits sounds out of that though. I donīt like the usual "both on" positions 2 and 4 as they are just not interresting and boring to me, so I went ahead and split them (gotta change the wiring on my les paul style already xD).
In my Ibanez S I have Mule neck, IT middle and RY bridge (pretty similar to your setup) and I made positions 2 and 4 to be a combination of middle and the outer coil (the slug coil here) of the bridge / neck PU.
While the top end does not work well with high gain, for low gain or clean they bring very interresting sounds, reminicent of tele sounds.
To me this is much more usable and different from the normal parallel aproach.
For this I used a RWRP middle, but after watching the comparison video put up a while ago I would have gone with a stock.
